Chromatin remodeling: Activating ACL1 with a little help from 'friends'

ScienceDaily (Nov. 29, 2012) ? Chromatin remodeling -- the packaging and unpackaging of genomic DNA and its associated proteins -- regulates a host of fundamental cellular processes including gene transcription, DNA repair, programmed cell death as well as cell fate. In their latest study, scientists at the Stowers Institute for Medical Research are continuing to unravel the finicky details of how these architectural alterations are controlled.

Through a series of biochemical experiments, Stowers Investigators Ron Conaway, Ph.D., and Joan Conaway, Ph.D., and their team discovered that chromatin remodeling enzyme and suspected oncogene ALC1 (short for Amplified in Liver Cancer 1) is activated through an unusual mechanism: Its shape shifts in the presence of its activators. Their finding identifies a new instrument in cells' molecular repertoire of chromatin-remodeling tools and a potential cancer therapeutic target.

One of the main tasks of chromatin remodeling enzymes, Ron Conaway explains, is "to make DNA accessible so events like repair and gene transcription can occur." Postdoctoral research associate and first author Aaron Gottschalk, Ph.D., previously figured out that ALC1 required protein partners to activate its remodeling function. Publishing in an upcoming issue of the Journal of Biological Chemistry, he dissects the mechanism by which this occurs.

ALC1 and its ilk have a common protein domain, SNF2, that uses the energy of ATP hydrolysis to move nucleosomes -- the basic repeating units of chromatin -- around, in a process called nucleosome sliding.

"I was intrigued because ALC1 has a unique macrodomain not found on any other SNF2 family member," Gottschalk says. His interest was further piqued when he found that while most ATP-dependent chromatin remodelers function as large multi-protein complexes, ALC1 appeared to work by itself. At the same time, where most of its group-happy family members readily demonstrated nucleosome sliding activity in vitro, ALC1 was not only a lone ranger but also "completely dead on its own."

Gottschalk deduced that ALC1 may function independently, but it needs a boost from a couple of sidekicks: PARP1, an enzyme that responds to several kinds of DNA damage; and NAD+, the substrate by which PARP1 transfers chains of poly (ADP-ribose) onto itself and other target proteins, in a process called PARylation.

Only when PARP1 and NAD+ are on the scene does ALC1 spring into action, altering the accessibility of DNA by shifting nucleosomes around. Gottschalk's earlier findings were published in the Proceedings of the National Academy of Sciences in June 2009.

"We then extended this research," Conaway says, "and the upshot of our recent JBC paper is that ALC1 is likely activated through a series of physical interactions. ALC1's unique macrodomain can bind PAR, and protein-protein interactions also occur between ALC1 and PARP1." Gottschalk and coauthor Rushi Trivedi, a graduate student in the Biochemistry & Molecular Biology department at KU Medical Center, developed a novel footprinting assay that enabled this observation. Rather than merely activating ALC1 and moving on, the researchers found that the trio of PARP1, NAD+ and ALC1 hangs out in a stable complex.

"So PARylated PARP1 and NAD+ are allosteric effectors -- by binding to ALC1, they alter its state from dormant to active," Conaway says. "It's an interesting mechanism that's different from how most other chromatin remodelers work. It may also help explain other evidence that PARP1 has the ability to rearrange nucleosomes and reorganize chromatin; this could be one way by which PARP1 exerts its influence."

Apart from its role in modifying chromatin structure, not much is currently known about ALC1. It's regarded as a possible oncogene, being found in excess in hepatocellular carcinoma cells and because overexpression of ALC1 in mice induces spontaneous tumors.

PARP1, on the other hand, has attracted plenty of interest as a potential anticancer drug target, due to its importance in maintaining genomic integrity. For example, in breast cancer cells lacking BRCA1 or BRCA2 function, blocking PARP could effectively remove the cells' last line of defense against DNA-damaging chemotherapy agents. To date, no PARP inhibitor has made it past phase III clinical trials, but pharmaceutical companies continue to chip away at the challenges around optimizing this form of targeted cancer therapy.

"A better understanding of the in-depth biochemistry we're uncovering on ALC1 and PARP1 may, in the long term, ultimately lead to new or more refined therapeutic strategies," Gottschalk says.

Meanwhile, having observed that ALC1 comes to life upon interacting with friends, Gottschalk now wants to understand precisely how this activation happens. With a knockout ALC1 mouse model handy, plans are afoot to extend his studies to an in vivo characterization of a chromatin remodeler that boasts an illustrious family pedigree, yet stands apart in the crowd.

Funding for this study came from a National Institute of General Medical Sciences grant (GM41628), and a grant to the Stowers Institute from the Helen Nelson Medical Research Fund at the Greater Kansas City Community Foundation.

TEXT-S&P Summary: First Ship Lease Trust

(The following statement was released by the rating agency)

23-Nov-2009 BB-/-- BB-/--

===============================================================================

Rationale

The rating on Singapore-based First Ship Lease Trust (FSL) reflects the

company's exposure to the weak credit profiles of its lessees and likely weak

freight rates over the next one to two years due to a downturn in the global

shipping industry. FSL's predictable cash flows from long-term lease contracts

protect the company from the volatility--to the extent its customers meet

their rent obligations. An improvement in FSL's short-term liquidity also

supports the rating.

We assess FSL's business risk profile as "weak." We expect conditions in the

shipping industry to remain difficult in 2013 due to an oversupply of ships,

tepid demand, and high bunker fuel prices. Over the past one to two years,

several FSL vessel charter contracts were terminated because the

counterparties faced financial difficulties. This increased FSL's exposure to

the spot market and decreased its EBITDA margin. In our view, a prolonged

downturn in the shipping industry could further increase the risk that some

more lessees may fail to pay rentals on time and renegotiate or terminate

charter arrangements. In our base case, we expect FSL's EBITDA margin to fall

below 70% in 2012 from 77% in 2011, and stay at those levels for one to two

years.

We assess FSL's financial risk profile as "aggressive." In our base-case

scenario, the company's key financial ratios are likely to remain weak. We

expect FSL's ratios of funds from operations (FFO) to debt and of debt to

EBITDA to be 11%-15% and 5x-6.5x, respectively, in the next one to two years.

The EBITDA interest coverage is likely to be 3x-4x in the same period. Most of

FSL's long-term contracts generate predictable cash flow. The company's

improved short-term liquidity following revisions in two loan covenants and

suspension of cash dividends until June 2013 support its financial risk

profile. However, we believe the downside risk to FSL's financial performance

is still significant given that counterparty risks are likely to remain high

in the next one to two years. Our base case does not incorporate potential

losses from further customer defaults or restructuring.

Liquidity

We assess FSL's liquidity to be "adequate," as defined in our criteria. The

company has no short-term debt maturities. FSL's liquidity profile has

improved following the relaxation in two covenants and the suspension of

dividend distribution until June 2013. We expect the ratio of liquidity

sources to uses to be 1.3x-1.4x in the next 12-18 months. Our liquidity

assessment is based on the following assumptions:

-- FSL's sources of funds include cash and cash equivalents of US$18.2

million as of Sept. 30, 2012, (after excluding US$15 million minimum cash

balance that the company is required to maintain as per its loan covenants)

and estimated FFO of about $50 million.

-- FSL has US$44 million of scheduled debt amortization per year and

scheduled interest payments of about US$24 million.

We assume that FSL will not acquire any new vessels in 2012 and 2013.

Outlook

The negative outlook reflects our view of the high credit risk of FSL's

lessees and the company's exposure to the volatile spot freight market amid a

prolonged downturn in the shipping industry.

We could lower the rating if FSL incurs large losses from defaults by

customers, or we see high likelihood of a covenant breach. In addition, lower

spot market freight rates than we expect could weaken the company's credit

protection measures and exert downward pressure on the rating. EBITDA coverage

of gross interest expenses of less than 2.5x or a ratio of FFO to gross debt

below 10% would indicate such weakening.

We could revise the outlook to stable if we see clear signs of an improvement

in the credit quality of FSL's lessees, along with a sustained improvement in

the company's credit protection measures and liquidity. Given that

counterparty risks are likely to remain significant over the next 12 months,

we believe the possibility of a positive rating action is low.

Direct Mail And Facebook For Website Traffic - Internet Marketing ...

If you?re looking to improve the amount of brand new customers that come into your business each and everyday, you will have to find new ways to promote yourself online, and do a wide variety of marketing strategies on a daily basis. If you want immediate sales, you have to be a traffic generation master.

Now getting traffic to your website doesn?t have to be hard. All you have to do is follow a few simple techniques that will allow you to have the most success as possible in your internet business. I?ve been doing this for a long time now, and I can honestly tell you that if you want to get more new sales, you have to drive more and more traffic to your website on a consistent basis.

In today?s lesson, I want to share with you some of the traffic methods that are working for me now, and how you can use these same techniques to work in your online business also. These strategies are very simple, and once you read them, you will see that it?s very easy to make money online simply and easily.

Here?s the first traffic method that you can use in your online business today:

1) Direct mail to existing customers

Once you develop a large group of customers, there will come a point in time where the traffic from your existing will bring you 80% of your total business profits. Because of this, you will want to do anything that you can to sell more and more of your products to these people - over and over again.

One of the best ways to increase your conversion results and traffic count is with direct mail. You see if you just rely on email marketing to contact your existing customers, you?re missing out on a great bunch of potential profits that you could be getting in your online business. Direct mail has been proven to increase sales of online business owners, and the same can be true of you if you decide to put it into use in your business.

Here?s another way to get traffic to your website:

2) Facebook

Facebook is the number 2 ranked website on the internet (in terms of website traffic), and they are right behind Google (Youtube is number 3 by the way). The more ?likes? and members that you can get to your Facebook page, and the more you interact with them, the more people will start to visit your site repeatedly to learn more free information or even to buy some of your products.

I think all businesses can benefit from Facebook. You can build alot of relationships with your Facebook members, and can create special deals, campaigns, and activities that will allow you to get more website traffic, and boost your sales? all for free. But the first step is getting those Facebook ?likes? and getting people to respond to you.

These 2 ways to get traffic to your website are both very good ways to make your business a success, and to start earning the money that you are looking for in your business. Hopefully you will put them into use.

Good luck with earning the money that you are looking for in your online business.

Nico Lang: 10 Love Lessons You Can Learn From the '80s

1. Dancing is the key to winning someone's heart. (Dirty Dancing, Flashdance, Footloose, Fame, Breakin' and Breakin' 2: Electric Boogaloo)

For anyone who has seen Dirty Dancing and the wonder of Patrick Swayze's torso (#neverforget), you know that nobody can resist dancing. What's sexier than someone who is in touch with their body and knows how to move it? It doesn't even matter what they're dancing to or whether it's a tune as admittedly dopey as "I've Had the Time of My Life." When you see those hips moving to Bill Medley and Jennifer Warnes' words, nothing else matters; not maudlin theme songs, botched backstreet abortions, the high expectations of your parents (Emily Gilmore and pre-L&O Jerry Orbach) or that you think you can't do that lift. You can do that lift. You just have to learn to believe in yourself. His sweet moves will show you the way.

Also, you know you kind of still love that song anyway.

2. Any problems you have with your romantic partner can be solved with the soulful tunes of Peter Gabriel. (Say Anything)

John Cusack movies taught me everything I needed to know about relationships, and Say Anything is a great example. Did you give her your heart and she gave you a pen? Do you want to be a professional kickboxer while she wants to study in England? Is her father, the dad from Frasier, being investigated by the IRS for tax evasions? All of these issues can be solved by blasting/serenading your beloved via boombox outside of her window. If you can't figure out what to say to win back her heart, let Peter do the talking. Like in 1977's Close Encounters of the Third Kind, music is the universal language, and when it comes to love, sometimes Gabriel's lyrics are the only words you need.

3. Hot guys have feelings, too. (Sixteen Candles, The Breakfast Club, Pretty in Pink)

Hey, just because he's beautiful, perfect and has cheekbones that could slice a diamond doesn't mean he isn't soulful or doesn't want a real relationship with someone. For example: You know all those douchebag friends of his that wreck his parents' house or that awful ex-girlfriend who let an unlicensed freshman operate his dad's Rolls Royce? He doesn't like those things, either. He might just like you and if you let him in, he will wait for you outside of your sister's wedding with a trans am, ready to do it on a cloud without getting pregnant or herpes. Or you'll just eat cake together, because who doesn't love cake? Either way, someone will remember your birthday and see how special you are, and it just might be him. So, when he tries to talk to you at the dance, make sure to say something back.

4. Sometimes the bad boy is just bad. (Heathers)

If the object of your affections is a dead ringer for Jack Nicholson in The Shining, has suspicious access to firearms and explosives and helps you plot to kill all of your friends, you might not want to take him to the prom. Before getting involved with the bad boy, ask yourself: "Does his father blow things up for a living? Did he move to your town under suspicious circumstances? Does he wear long trenchcoats all the time? Is he anti-social or a possible serial murderer?" If you are unclear about the answers to any of these questions, run. "Ich Luge" bullets are not a thing -- you're the one who is lying to yourself.

5. Opposites attract. (Pretty in Pink, Footloose, every '80s movie ever)

Just because he's popular doesn't mean he's not into you. You're the girl from the wrong side of the tracks (who has to make her own dress for the prom), the sort-of-goth girl who needs someone to open up to or the preacher's daughter who just wants to let her wild side out and dance but can't because the town outlawed dancing. Although your families don't approve and it seems like you couldn't possibly get together with society standing in the way, nothing is so insurmountable that it can't be solved before the closing credits. You'll get a makeover, find your self-esteem and get your dad to sit down and listen to what the kids are jamming to these days. When John Lithgow finally hears Kenny Loggins, he'll understand what Paula Abdul and a cartoon cat have been telling us all these years: Sometimes it's so wrong that it couldn't be more right.

6. Want to get the guy? Be yourself. (Sixteen Candles)

This is the '80s, and girls aren't like Sandy in Grease, ditching their poodle skirts to impress some guy. No. You're like Molly Ringwald, and guys like you for just doing you. You're not like other girls. You wear kooky-yet-cool flowered hats and loan your underwear to a freshman, because you're special and you care about people. If he really likes you, he won't care about his popular friends or that you have an overtly racist Asian caricature staying in your house. When you like someone, you like them for the quirks that make them who they are, even if those quirks are Long Duk Dong.

7. I repeat: Don't change for them. (The Breakfast Club, Pretty in Pink)

'80s movies are big on transformations. We find out that nerdy guy was McDreamy THE WHOLE TIME or that Ally Sheedy is a knock-out when her bangs aren't covering up her entire face. Getting a new wardrobe and a new lease on self-esteem is wonderful, but don't let someone else tell you who to be. A makeover can turn a frog into a prince, but that only goes so far. When Patrick Dempsey gets the girl in Can't Buy Me Love, it's because he's been a prince THE WHOLE TIME. Change the clothes, but don't forget about the person inside them.

8. Guys who ride riding mowers are surprisingly sexy. (Can't Buy Me Love)

Although country music has informed us of the erotic powers of the tractor, CBML makes a strong case for the sexual appeal of lawn trimmers. However, it helps when the person riding that thing is as forever-hot as Patrick Dempsey, who only seems to get more good looking with each passing year. Between him, Robert Downey Jr. and Anthony Michael Hall (who got weird hot after the '80s), I'm convinced there's a Brat Pack Fountain of Sexy, and I want in. At the very least, they need to share with Judd Nelson.

9. Friendship is the basis of any great relationship. (Some Kind of Wonderful, The Goonies)

Although the two of you CAN just be friends, sometimes being best friends with someone can be the foundation for something more. When Harry Met Sally is the classic example of this (because who wouldn't eventually fall in love with Meg Ryan?), but Some Kind of Wonderful proves this conclusively. You seem like bros, but this just masks the fact that you've got it for him, bad, even though he has the hots for Lea Thompson.

However, he will eventually watch Howard The Duck, where she has intergalactic nookie with a cigar-smoking alien waterfowl, and he'll get over her. Guess who will still be there? You. You put in the time and the effort and were there for him while he got his heart broken by poultry porn, and that matters. Sure, you might have to chauffeur him on a couple dates to get to that point, but when he sees how cute you look in that chauffeur hat, he'll never think about Lea Thompson again.

10. But sometimes, you have to settle for being friends. (Pretty in Pink)

Sure, you could end up in a Beauty and the Beast (1991) situation, where there's something there that wasn't there before. But if you're pining for her and she makes it clear she wants to be with the angelicly feather-haired Andrew McCarthy, you need to give it up, bra. You can best win her over by being there for her when Andrew's bougie best friends are a jerk to her for being, like, poor and stuff. (Dem blue collars are such a buzzkill, amiright?) Either she'll find happiness with Andrew McCarthy when he finally gets up the courage to tell off James Spader and you'll come to respect her decision, or you'll forge an alternate reality where a Flock-of-Seagull-haired Jon Cryer can find love, too. Maybe that will then stop him from doing Two and a Half Men? A girl can dream.

One of FBI's most-wanted held in Mexico

By NBC News staff and wire reports

One of the FBI's Ten Most Wanted Fugitives was arrested in Mexico and returned to Los Angeles Friday night to face charges of murder, kidnapping and rape, U.S. officials said.

Reputed Los Angeles gang member Joe Luis Saenz was taken into custody in Guadalajara late Thursday following a joint operation with the Mexican government, Bill Lewis, assistant director in charge of the FBI's Los Angeles office, said.

Investigators said Saenz shot and killed two rival gang members in July 1998 to retaliate for an assault on one of his associates.

Saenz suspected Sigrieta Hernandez, his girlfriend and the mother of his daughter, was going to tell police about the slayings, investigators said.

He is accused of kidnapping, raping and killing her less than two weeks later.

Videotape murder
Saenz also is believed to have killed Oscar Torres at his home in suburban Whittier in October 2008 because he failed to repay $600,000 in drug money after police seized the cash during a traffic stop.

Authorities said they have videotape from a surveillance camera at Torres' house that shows Saenz killing Torres and wounding another person.

Saenz was still listed on the FBI?s most-wanted list early Saturday, but with a red caption on his photograph reading ?CAPTURED.?

Born in Los Angeles, Saenz was known to travel between the United States and Mexico.

Saenz, who is about 37 years old, was believed to be hiding in Mexico, working as an enforcer and hit man for a Mexican drug cartel.

He had a number of aliases including Zapp, Peanut Joe Smiley and Honeycutt, it added.

Saenz had been on the FBI's most-wanted list since 2009, putting him among the ranks of Osama bin Laden, Boston crime lord James "Whitey" Bulger and other notorious criminals.

There was a reward of up to $100,000 for information leading to his arrest.

The Associated Press, Reuters and NBC's Ian Johnston contributed to this report.

Bolt, Felix named IAAF World Athletes of the Year

BARCELONA, Spain (AP) ? Usain Bolt won the IAAF World Athlete of the Year award for the fourth time Saturday after defending his 100 and 200 meter gold medals at last summer's Olympic Games in London.

The Jamaican sprinter, who beat out American hurdler Aries Merritt and Kenyan runner David Rudisha, had previously won the award in 2008, 2009, and 2011.

"For me this is a great honor to win a fourth time. I really worked hard and I was really focused this year. This season was one of my toughest. I had my ups and downs, even though we don't like to talk about them," said Bolt, who thanked his coach and the fans at the Olympics.

"The London games were magnificent. Fans were there in the stadium from 10 in the morning. I had never seen anything like that."

Sprinter Allyson Felix of the United States took home the women's award.

In 2003, Bolt and Felix won the Rising Star awards for the most promising young athletes of the year.

"Nine years ago I was here with Allyson, now I am back, that means I am getting old, which is slightly worrying," said the 26-year-old Bolt. "But I will continue to work hard."

Felix won the 200-meter gold medal in London after settling for silver four years earlier in Beijing.

"My biggest defeats have come at the Olympic Games. After Beijing I was devastated, I had worked as hard as possible but I came up short," she said. "But I decided to rededicate myself even harder and it was a blessing in disguise. I was able to work harder than ever before and finally it all came together."

The other finalists for the women's award were New Zealand shot putter Valerie Adams and British heptathlete Jessica Ennis ? both gold medalists in London.

Merritt won the Inspiration Award for his season, which included a gold medal and a world record in the 110-meter hurdles.

Rudisha, who was named Athlete of the Year in 2010, won the Performance of the Year award for his record-setting gold medal run in the 800 meters.

Javelin thrower Keshorn Walcott of Trinidad and Bahamas sprinter Anthonique Strachan won the Rising Star awards.

The awards were announced on the final day of the IAAF's 100th anniversary celebration.

Va. center aims to help US reclaim manufacturing

DISPUTANTA, Va. (AP) ? Tucked away just miles from the railroads that for years have transported goods made in Virginia to the rest of the world, a recently opened research facility in Prince George County is bringing together universities and industry in an effort to help the state ? and the country ? regain its manufacturing roots.

The work being done at the Commonwealth Center for Advanced Manufacturing will be used for production that has come a long way from the textiles and furniture plants that once populated southwest and Southside Virginia. The targets now are jet engines, laser printer cartridges and military submarines.

"Most people's image of a manufacturing facility is one of a hot, dirty, undesirable environment in which to work. Historically that may have been the case, but in today's modern factory, that is in fact exactly the opposite of what it is," said Dave Lohr, executive director of the 62,000-square-foot facility, which features natural lighting, streamlined glass and metal with a sheen like that of the latest electronic gadgets.

With office space, meeting rooms, labs and a large, cavernous area to house and test the latest high-tech equipment, the center completed in September is conducting research for a group of manufacturing companies under a partnership with Virginia Tech, Virginia State University and the University of Virginia. The consortium supporting the center's research through equipment, funding and expertise includes Rolls-Royce, Cannon, Newport News Shipbuilding, Siemens and others.

Much of the research that will be done at CCAM focuses on surface engineering and advanced products and components, as well as manufacturing automation and simulation.

The center is currently involved in 10 projects valued at $1.7 million that includes both generic research that could apply to many of the 14 industry members, and direct research that addresses specific topics raised by one or more of the companies. Lohr said he hopes the center ultimately will be involved with between $10 and $15 million in research each year.

One of the first projects being conducted at the center looks at improving the technology used to coat surfaces with anti-heat or friction material using a robotic arm that can spray those coatings on metals. Another is studying human performance in manufacturing operations. As part of those research efforts, the center also will have a visualization lab that will allow engineers to do three-dimensional modeling.

CCAM is a "cornerstone of innovation" that "provides a place for researchers and industry partners to collaborate on the innovative technologies of tomorrow," said Will Powers, executive vice president and CFO of Rolls-Royce North America, which donated the land for the building on its Crosspointe campus, where it makes components for jet engines.

Construction of the building, which is owned by the University of Virginia Foundation, was financed with $11 million from state Recovery Act bonds, a $4 million federal grant and a $2.5 million grant from the Virginia Tobacco Indemnification and Community Revitalization Commission.

Those involved say the center allows industry and universities to solve real problems and gives companies a low-risk environment in which to examine new ideas.

"The factory floor today isn't designed for experimentation, it's designed for efficient production. When you're interrupting that with experiments, then you disrupt the intended purpose of the factory," Lohr said. "Having an independent location where you can do research and do it at the same speed as you would at a factory, it takes that risk out of the equation."

Lohr said when you're making really expensive parts, "you don't want to send one to the lab and cut it up after you've invested all the money making it ? you want to sell it."

Similar public-private collaborations are taking place across the world.

In the U.S., BMW partnered with Clemson University in Greenville, S.C., for research that focuses on automotive technology, Lohr said. And President Barack Obama announced a $30 million federal investment in a manufacturing innovation institute in Youngstown, Ohio that will bring together manufacturing firms, universities, community colleges, and non-profit organizations from Ohio, Pennsylvania and West Virginia.

From a workforce standpoint, Barry Johnson, senior associate dean at the University of Virginia's School of Engineering and Applied Science, said the collaboration allows universities to better understand what types of jobs and skills needed in the advanced manufacturing workplace, ultimately putting the U.S. in a better position to attract factories that may have otherwise gone to areas overseas.

"Companies have the ability to put these businesses anywhere in the world, and they're going to put them where those factors are favorable to them," Johnson said. "We in the U.S., we can compete with China, we can compete with India, we can compete with other parts of the world that have low labor rates."

Lohr agrees.

"The opportunity to reclaim manufacturing as a viable industry in the United States is there for the taking, if you do it with advanced manufacturing processes," he said. "While it may not create jobs as you might have had in a classic factory, what it is creating is a very high-value job."

Which 'habits' influenced you most? ? Business Management Daily ...

Stephen Covey, author of the landmark self-help book The Seven Habits of Highly Effective People, passed away July 16.

Scott Eblin, a leadership coach, shared his reflections on his favorites of the habits.

???Begin with the end in mind. This is especially important for long-term goals. ?By their nature, longer term goals are usually less clear and the path to achieving them is less certain. I?m still learning how to articulate them and then reverse engineer back from those goals in bite-size chunks that are actionable in the short run,? Eblin writes.

???Put first things first. ?With email overload and 24/7 connectivity via smartphone, the world is a lot different today than it was when The Seven Habits was first published in 1989,? Eblin writes. ?Technology makes it much easier for others to impose their urgencies upon you today than 23 years ago.?

???Seek first to understand, then be understood. ?A few years ago I learned that this habit was first articulated by St. Francis of Assisi. Props to Stephen Covey for bringing it back to modern day attention,? Eblin writes.

Nicaragua orders China the construction of a satellite | NexTV

The Nicaraguan Government has formalized the acquisition of a communications satellite for USD 244 million, with the Chinese firm Gran Muralla China. The latter will start with the construction of Nicasat-1 in January 2013 and it is expected to be placed in orbit by mid-2015. The Nicaraguan Pay-TV market is currently led by the cable and DTH operator Claro TV (Am?rica M?vil), followed by Sky (DirecTV/Televisa), and cable operators IBW and Telecable.

The agreement was executed in October by representatives of the regulating entity of the country and Instituto Nicarag?ense de Telecomunicaciones (Telcor) and the Chinese Gran Muralla China. Such agreement was entered into after several months of negotiations in which the country managed to lower the satellite construction Price from USD 300 million to USD 244 million. ?Funds are guaranteed but the loan has not been executed yet?, stated Telcor President, Orlando Castillo.

Nicasat-1 will render satellite services to Nicaraguan communication and television firms and other Central American countries. This project will enable savings for USD 8 million annually, which is the amount currently paid by telecommunications firms for the use of such services. Additionally, cellular telephony, Internet access and Pay-TV prices are expected to be reduced, thus enabling the service arrival in rural areas where cable does not operate.

Two powers, Qatar and Iran, try to sway Hamas

(AP) ? The courtship of Hamas between rivals Iran and Qatar has been one of the Middle East's intriguing subplots of the Arab Spring. The bloodshed in Gaza has now sharpened their competition for influence with the Palestinian militant group and the direction it takes in the future.

Qatar has sought to use its vast wealth to win over Hamas with investments and humanitarian aid and encouraging Arab partners to do the same ? part of the hyper-rich U.S. allied nation's broader campaign to bring under its wing Islamist movements that have risen to power in the region the past two years. Qatar's influence with Hamas could edge it away from armed action toward diplomacy.

Iran, meanwhile, is invigorating its longtime role as the builder of the rocket arsenal for Hamas' military wing.

For Hamas, there are benefits in both directions ? and it's happy to play both sides. During a celebration rally in Gaza City after an Egyptian-mediated cease-fire came into place ending fighting between Israel and Hamas, Gazans wildly waved flags of Qatar, along with those of Egypt and Turkey, in gratitude for those countries' diplomatic support.

At the same time, Hamas' leader-in-exile Khaled Mashaal, who is based out of Qatar, gave a very public thanks to Iran for standing by Gaza with crucial military assistance. Fighters in Gaza also hailed the new reach of their arsenal, with Iranian-designed Fajr-5 rattling Israel by reaching the outskirts of Tel Aviv and Jerusalem.

Visiting the Syrian capital Damascus on Friday, Iran's parliament speaker Ali Larijani, who is close to the country's supreme leader, promised leaders of Palestinian militant groups that his country would continue to boost "the resistance's capabilities in confronting the Zionist arrogance and aggression," according to Palestinian official Khaled Abdul-Hamid, who attended the meeting.

The reminder of Hamas' reliance on Iran for weapons could help smooth a relationship that has been running through a rough patch because of the civil war in Syria, Iran's top ally.

Embarrassed by the Syrian regime's crackdown on a mainly Sunni Muslim uprising, Hamas leaders based in Damascus for years broke with Syria and left for Qatar and Egypt. Though Iran continued to send weapons to Hamas, the break undermined the "Axis of Resistance" grouping Syria, Hezbollah and Hamas that Iran has assembled in the Arab world.

It's doubtful Iran can fully reclaim its position as the main big brother for Hamas. But Tehran's image is certain to receive some lingering boost in Gaza.

For Hamas, hyper-rich Qatar is a political and economic lifeline, a key part of the militant group's attempts to bolster its ties with the Western-backed Gulf states in efforts gain more international legitimacy. Last month, Qatar's emir became the first head of state to visit the Gaza Strip since Hamas took control five years ago. The Gulf state pledged nearly $500 million in aid and a song called "Thank you, Qatar" played on Gaza radio and TV as the emir was given a hero's welcome.

During the heat of the Gaza battle the past week, Qatar's prime minister gave a blistering dressing down to the Arab League during an emergency meeting, saying Arab nations had to do more to fight Gaza's poverty and isolation than just pass resolutions.

"We can't give hope without delivering," Sheik Hamad Bin Jassem Al Thani told the gathering last week in Cairo.

Two days after his outburst at the Arab League meeting in Cairo, Sheik Hamad suggested that his country would be willing to open dialogue with Israel on a long-term Gaza truce if it leads to lifting the blockade. The Arab Spring, he added, has made Israel feel more vulnerable, but also perhaps more ready to make deals.

"We need to talk with everyone to reach a comprehensive peace," he told CNN on Monday.

For Qatar, the outreach to Gaza also is part of far wider ambitions to become a major policy-shaper in the Middle East. The tiny Gulf nation has emerged as a strong backer of the Muslim Brotherhood, which has risen to power in Tunisia and Egypt after the fall of those countries' autocratic leaders in early 2011.

In February, Qatar brokered talks between Hamas' Mashaal and his longtime rival, internationally backed Palestinian President Mahmoud Abbas.

Qatar has also sought to influence Syria's rebels. This month, it hosted Syrian opposition groups in a breakthrough effort to unite rebel factions under one coalition, which has opened the way for greater international recognition and promises of aid. Qatar had led calls to supply Syrian rebels with heavy weapons to counter air and tank attacks by Bashar Assad's forces. Qatar also was a key backer of the Libyan uprising.

On Thursday, Qatar also invited the newly formed Syrian opposition coalition to appoint its ambassador to the Gulf state, the Qatari news agency reported.

Salman Shaikh, director of The Brookings Doha Center in Qatar, said it's not a matter of "having to go all for Iran or all for Qatar.'

"What Qatar is trying to do is change the reality. They are trying to blaze a trail that will weaken the international isolation of Gaza from the Israeli blockade," he said.

Qatar has so far stopped short of offering any kind of military support for Gaza to avoid a rift with Washington, Israel's most powerful ally.

Iran takes a very different view.

Officials in Tehran boasted this week about its longtime arms support for Hamas militants ? part of Iran's bookend strategy to equip anti-Israel factions in Gaza and Hezbollah in Lebanon on Israel's northern border.

Gen. Mohammad Ali Jafari, the head of the Revolutionary Guard, said Iran supplied fighters in Gaza with the technology to "quickly" produce the Fajr-5 missiles. The statement fits with previous Iranian denials that it is not directly sending missiles to Gaza, but suggests close coordination on construction and movement of supplies, presumably through the smugglers' tunnels linking Gaza and Egypt.

The Gaza fighting, at the least, bought Iran some restored street credibility as its image was battered by its backing for the regime of Syrian President Bashar Assad.

"The war with Israel reminded Hamas that Israel is the main issue not Syria," said Heshmatollah Falahatpisheh, a professor in politics in Tehran's Allameh University.

But Hamid Reza Shokouhi, editor of Iran's independent Mardomsalari daily, said the bump in Iran's popularity with Gazans could be only "short-term." He questioned whether Iran could sustain its influence in Gaza against the almost unlimited resources of Qatar and its Gulf partners.

Iran's Foreign Ministry and a parliamentary committee have applied for permission to visit Gaza in the coming weeks via the border crossing with Egypt, said Hasan Qashqavi, deputy foreign minister in charge of consular affairs.

Qatar's prime minister said the competition for influence is Gaza only likely to intensify as other nations such as Turkey and Egypt reach out.

"We are not trying to take Hamas from anybody else, from Iran or others," he said in the CNN interview. "Hamas, they have to decide for themselves. I think they are pretty mature to decide for themselves."

Arthritis National Research Foundation and American Federation for Aging Research collaborate to support research on arthritis and aging

The Arthritis National Research Foundation (ANRF) in Long Beach, CA and the American Federation for Aging Research (AFAR) in New York, NY proudly announce their collaboration to fund an Arthritis and Aging Research Grant. The Arthritis and Aging Research Grant provides up to $100,000 for one year to junior faculty studying the role of aging in the development of arthritis. A second year of funding may be available if significant progress is demonstrated. The grant application process is now open for 2013.

"This collaboration with the American Federation for Aging Research enables us to pool our resources and relationships within the scientific community to expand our outreach and opportunity for new knowledge in the field," said Arthritis National Research Foundation executive director Helene Belisle.

The Arthritis National Research Foundation (ANRF) is the charity that funds research to cure arthritis.

"We are pleased to be collaborating with the Arthritis National Research Foundation. Understanding the connections between the fundamental mechanisms of aging and arthritis will move us closer to effective treatments and cures for arthritis," said Stephanie Lederman, executive director of AFAR.

Aging is a major risk factor for some forms of arthritis. Osteoarthritis for example, is strongly linked to aging but the mechanisms for this link are incompletely understood. With this grant, AFAR and ANRF aim to support new research in this still underexplored area. "We are enthusiastic about this partnership; it is a perfect fit for both organizations," Belisle added.

Applications for the research program must be submitted online, at the ANRF website, no later than January 18, 2013, at 5:00 pm PST. The grant period is from June 1, 2013 to May 31, 2014.

"Our goal is to cure arthritis for people suffering worldwide," added Belisle. "With our aging population, this disease will be increasingly prevalent. Research funded by the collaboration of ANRF and AFAR could uncover new information to help find a cure."
